1. The gene - Enzymatic manipulation of DNA - Cloning - Vectors used in prokaryotes - Bacterial hosts


2. Heterologous expression in Gram-negative bacteria - Production of recombinant proteins and gene fusions


3. Heterologous expression in Gram-positive bacteria - Molecular biology and genetic manipulation of yeasts.



4. Cosmids, phagemids and phage display, methodologies and applications.



5. Mutagenesis techniques and their applications - Site-targeted mutagenesis - Saturation mutagenesis - Directed evolution.



6. Plant genetic engineering: transformation methods - Agrobacterium tumefaciens - Stable transgenesis & agroinfiltration - Application examples.



7. Expression of proteins in mammalian cells - Transfection techniques - Vector viruses - siRNA / shRNA.



8. Detection of protein-protein interactions (Two-hybrid system).

 

9. Genome editing / Transgenesis.

 

10. Genome editing / Transgenesis (applications).

Learning outcomes of the learning unit

· To get acquainted with the main methods used in genetic engineering
· To get informed of the potential of genetic engineering in the study of living organisms, as well as in the improvement of species for human use and for the production of recombinant proteins.

Prerequisite knowledge and skills

Molecular biology and genetics (courses of the bachelier in Biology).

This course concerns only students who attended it but did not pass the exam during the academic year 2022-2023. From 2023-2024, there will be no face-to-face lectures. Questions/answers sessions will be organized on request.
