| Course contents : | Partim Structure. The different experimental steps that will lead to the image of a biomolecule using the technique of X-ray diffractionby monocrystal will be described. Three examples illustrating the relations structure-function will be approached, on the basis of research articles describing structural analysis of different proteins illustrating the following 3 families: - proteins controlling the DNA transcription, - serine proteases, - membrane proteins.
Partim Dynamic. This series of lectures is based on the use of biophysical methods (mainly optical spectroscopies, presented into details for course CHIM9218-1 : Biospectroscopies et résonance magnétique nucléaire appliquée à la biologie) to study protein folding. The optical properties (i.e. absorbance, fluorescence, circular dichroism and infrared) of proteins will first be reviewed, followed by the analysis of concrete examples (e.g. bêta-lactamases, single-domain antibody fragments, lysozyme) found in the literature. |

| Course objective : | Partim Structure. This course gives to the future biochemists the bases necessary to understand the principles of determination of 3D structures by X-ray crystallography in order to enable them to make an objective use of the crystallographic models.
Partim Dynamic. Give insights into the use of biophysical methods to study protein folding. |

| Prerequisites : | Partim Structure. Course of physics of 1st cycle. Basic knowledge in X-ray crystallography and knowledge of the basic principles of the structures of proteins.

| Workshops : | Partim structure and dynamic. This lecture will be given in a classical form, including the use of a PowerPoint presentation, a chalk and a black board. |

| Assessment : | A written exam will be organized for each part of the course and it will be compulsory to obtain at least 8/20 for each of them. |
