Course contents :  Structure part (20h+20h):
X-rays: A rapid overflight of the various necessary steps that will lead to the image of a biomolecule is carried out in the first 4 hours of theoretical course. Three examples illustrating the relations structure-function will be approached in the 6 last hours of theoretical course: - proteins controlling the DNA transcription, - serine proteases, - membrane proteins.
The 10 hours of TP will be dedicated to the analysis and the discussion of articles of structure concerning the 3 families of proteins described.

Dynamics part
The lecture begins with a short review of the spectral transitions. Afterwards, the following spectroscopic techniques are studied in detail : uv-visible absorption, linear and circular dichrosms, fluorescence and its polarisation, I.R. spectroscopy and surface plasmonic resonance. For each technqiue, experimental aspects and applications to the study of protein and nucleic acid conformations and of their interactions with small ligands will be developed
Course objective :  Structure part (20h+20h):
X-rays: This course gives to the future biochemists the bases necessary to understand the principles of determination of 3D structure by X-ray crystallography in order to enable them to make an objective use of the crystallographic models.

Dynamics part
Increase the qualifications of the student concerning the spectroscopic methods available for the study of the conformational dynamics of proteins (folding and unfolding), nucleic acids and their interactions.
Prerequisites :  Structure part : Course of physics of 1st cycle.
Xrays: Basic knowledge in X-ray crystallography (2° candidature in Biology or Chemistry) and knowledge of the basic principles of the structures of proteins (1st Licence in Biochemistry).

Chemistry of Biological Macromolecules Functional properties of biological macromolecules
Organization :  Structure Part:
Xrays:
Theory: 4 hours dedicated to the determination of the biomolecules structures by X-ray crystallography - 6 hours dedicated to the illustration structure-function with 3 examples.
TP: Analysis and critical discussion of articles in relation to the examples of structures described.

Dynamics part :
This lecture will be given as a classical form (as interactive as possible) by the two teachers according to their specific qualifications

Assessment :  Le mode d'évaluation sera choisi en concertation avec les étudiants. La réussite du cours (i.e. comprenant toutes ses parties) ne sera possible qu'à la condition d'obtenir au moins 8/20 auprès de chaque enseignant
