GENE0201-1

Genetics

Duration :25h Th, 15h Pr
Credits/ECTS :
2nd "candidature" in biological sciences4
required preliminary complement to register in the "licence" in biochemical sciences4
required preliminary complement to register in the "licence" in biological sciences4
Holder(s) :Claire Remacle
Course contents : The theoretical course is divided into 9 chapters. The first two chapters are devoted to the organization of genes and genomes and to their replication and expression (transcription, translation). The three next chapters concern gene transmission and recombination. Chapters 6 and 7 describe the types of mutations. Chapter 8 describes genetics of phages and bacteria. The last chapter briefly describes the technology of recombinant DNA.
Course objective : description of the basis of formal and molecular genetics
Workshops : Practical courses: one afternoon is devoted to bacterial conjugation in Escherichia coli (interrupted conjugation) and the other one to intragenic complementation of arg- markers in the unicellular green alga Chlamydomonas reinhardtii.
Bacterial conjugation: conjugation is made between a donor strain harboring a fertility plasmid and a recipient strain. Conjugation is interrupted at defined times. Transfer of plasmid is detected by plating bacteria on growth media supplemented with antibiotics.
Intragenic complementation: activity of the argininosuccinate lyase is measured before and after a heat shock in a wild-type haploid strain, in mutant haploid arg- strain and in a diploid strain showing intragenic complementation at the arg7 locus.
At the end of the theoretical course, one afternoon is devoted to the resolution of problems of genetics.
Written notes : Syllabus and Power Point slides are given to the students.
